摘要:观察感染后肠易激综合征小鼠肠组织中IL-17、occludin和ZO-1的动态改变。40只C57BL/6小鼠随机分为对照组、感染组(感染后2、4、6、8周组)。感染组小鼠用0.2mL含~条旋毛虫的生理盐水灌胃。于感染后2、4、6、8周,记录小鼠体质量,检测腹壁撤退反射(AWR),连续8h收集小鼠粪便,计算粪便含水百分数。HE染色观察肠道病理改变,免疫组织化学法及Westernblotting检测回盲部和结肠中IL-17、occludin和ZO-1的表达。感染后第2周小鼠肠道出现急性炎症反应,体质量显著降低(P=0.);至第8周肠道炎症和小鼠体质量基本恢复至正常。结直肠扩张容量为0.35和0.5mL时,感染组AWR评分均高于对照组(P0.01)。感染组粪便含水百分数均高于对照组(P0.05)。免疫组化及Westernblotting结果显示:与对照组相比,感染后2周组IL-17降低(P0.05),感染后8周组IL-17增高(P0.05);感染组occludin和ZO-1均低于对照组(P0.05)。IL-17的动态改变及紧密连接蛋白occludin和ZO-1的减少可能是导致PI-IBS小鼠内脏高敏感性和粪便含水百分数增高的原因之一,参与了PI-IBS的发生。

Abstract::ToobservethedynamicchangesofintestinalIL-17,occludin,andZO-1inmicewithpost-infectiousirritablebowelsyndrome(PI-IBS).FortyC57BL/6micewererandomlydividedinto5groups:controlgroupandinfectiongroups(2weeks,4weeks,6weeks,and8weeksaftertrichinellainfection).Infectiongroupsweregivenbygavagingof~Trichinellaspiralisin0.2mLofnormalsaline.Thebodyweightofmicewererecordedatweek2,4,6,and8afterinfection.Thevisceralsensitivityofmicewasmeasuredbytheabdominalwithdrawalreflex(AWR).Thestoolwascollectedcontinuouslyfor8hourstocalculatethepercentageoffecalwatercontent.PathologicalchangesofgutwereobservedbyHEstaining.TheexpressionsofIL-17,occludin,andZO-1inileocecusandcolonweredetectedbyimmunohistochemistryandWesternblotting.Atweek2afterinfection,theacuteinflammationoftheintestinaltractwasobservedandthebodyweightofmiceweresignificantlydecreased(P=0.).Untilweek8afterinfection,theintestinalinflammationandbodyweightofmicerecoveredtonormal.Whenthecolorectaldilatationcapacitywas0.35and0.5mL,theAWRscoresintheinfectiongroupsweresignificantlyhigherthanthoseinthecontrolgroup(P0.01).Thepercentagesoffecalwatercontentintheinfectiongroupsweresignificantlyhigherthanthoseinthecontrolgroup(P0.05).Comparedwiththecontrolgroup,theexpressionsofIL-17weresignificantlydecreasedinweek2group(P0.05)andincreasedinweek8group(P0.05).TheexpressionsofoccludinandZO-1intheinfectiongroupsweresignificantlylowerthanthoseinthecontrolgroup(P0.05).ThedynamicchangesofIL-17andthedecreaseofTightjunctionproteinsmaybeoneofthemechanismsofvisceralhypersensitivityandincreasedper?centagesoffecalwatercontent.TheymaybeinvolvedinthedevelopmentofPI-IBS.
